6-CF Uptake (Image based) 6-CF Uptake (image) Uptake of 6-CF (OAT Substrate), quantified by fluorescent image analysis
CD36+ monocytes CD36+Mono Quantification of mean fluorescence intensity (MFI) per cell of CD36+ monocytes (green signal) based on the images
CD80 + monocytes CD80+ Quantification of mean fluorescence intensity (MFI) per cell of CD80 + monocytes (green signal) based on the images
Cell Proliferation Proliferation Cell proliferation is the process that results in an increase of the number of cells, and is defined by the balance between cell divisions and cell loss through cell death or differentiation. [Nature]
Diameter Change Vasoactivity The vasoactivity of TEBVs was evaluated by placing the TEBV chip under a stereoscope (Amscope, software “ISCapture v3.6”) at 9 × amplification, recording an image of the initial outer diameter, and adding 1 μM phenylephrine (Sigma) to the flow circuit. After 5 min, the outer diameter was recorded and acetylcholine (Sigma) was added to the perfusion media for 5 min at a final concentration of 1 μM to assess vasodilation. The outer diameter change of the TEBVs was analyzed using Image J (NIH)
EAM-1 Uptake (image based) EAM-1 Uptake (image) Uptake of EAM-1 (OCT substrate), quantified by fluorescent image analysis
EC Alignment Alignment ECs (endothelial cells) align under perfusion compared to static culture
EC Aspect Ratio EC Elongation ECs (endothelial cells) elongate under perfusion compared to static culture
Hepatocyte Nuclear Factor 4, Alpha HNF4A The protein encoded by this gene is a nuclear transcription factor which binds DNA as a homodimer. The encoded protein controls the expression of several genes, including hepatocyte nuclear factor 1 alpha, a transcription factor which regulates the expression of several hepatic genes. This gene may play a role in development of the liver, kidney, and intestines. Mutations in this gene have been associated with monogenic autosomal dominant non-insulin-dependent diabetes mellitus type I. Alternative splicing of this gene results in multiple transcript variants encoding several different isoforms. (provided by RefSeq, Apr 2012)

Permeability - 10 μg/ml of FITC-IgG 10 μg/ml of FITC-IgG Serial fluorescence images with the same exposure time were taken every 5 min for up to 20 min. Then the permeability of TEBVs was calculated using equation: P= r/2ΔI * (dI/dt)t=0, where ΔI is the change in total fluorescence intensity upon addition of labeled molecule (t = 0)–Background signal; (dI/dt)t=0 is the initial rate of transport of fluorescent dextran from the vessel lumen into the vessel, which can be approximated as (dI/dt)t=0≈(It−I(t=0))/t, where It is the intensity outside the vessel at time t, It=0 is the intensity outside the vessel at time t = 0; r is the radius of the lumen, which is estimated from width/2 of the fluorescent region at t = 0.
